Not a whole lot. It's pretty much a single player experience with multiplayer interjections. You'll lose the ability to seek phantom assistance, and you won't get hints littered on the floor. On the other hand, you won't be invaded and you won't get misled by bogus hints.
I think, barring a sudden change of heart, I'll pick it up today, since a) I'm going to be in the vicinity of a Best Buy in an hour or so, b)I just remembered that I have a bunch of gift cards left over from Christmas, c) it's down to 40 bucks, and d) I'm feeling particularly masochistic today.
